New Plasmon Waveguides Composed of Twin Metal Wedges with a Nano Gap

We have numerically shown the existence of coupled wedge plasmons (CWPs) which propagates along a nano gap of a twin metal wedge. The CWPs are formed by wedge plasmons which can interact with each other. The dispersion relations of the wavenumber, propagation distance, and beam area of CWPs, are described and show that the characteristics of CWPs are similar to those of wedge plasmons and of gap plasmons. We also propose a new plasmon waveguide composed of twin metal wedges with a nano gap.     

Direct determination of benzamides in serum by column-switching high-performance liquid chromatography.

A column-switching high-performance liquid chromatographic method with fluorescence detection was developed for the simultaneous determination of four benzamide-type anti-psychotic drugs: sulpiride, tiapride, sultopride and metoclopramide in human serum. In this method, a TSKgel Super-ODS column was used as an analytical column, and a TSKgel G 2000SW was prepared as a pretreatment column. Under the optimized analytical conditions, four benzamide-type anti-psychotic drugs were eluted within 18 min. The detection limits (S/N = 3) for sulpiride, tiapride, sultopride and metoclopramide are 1 ng/ml, 4 ng/ml, 2 ng/ml and 0.5 ng/ml, respectively. Finally, the method was applied to the determination of sulpiride in human serum samples obtained after a single oral dose of sulpiride.     

Stereochemistry of cholesterol hydroxylation at C-22 during pregnenolone biosynthesis

Cholesterol and 20-hydroxycholesterol labelled at C-22 stereospecifically by deuterium were incubated with the enzyme of bovine adrenocortical mitochondria. The results indicated that hydroxylation occurs with retention of the configuration of C-22 to give exclusively the 22R stereoisomers of 22-hydroxy- and/or 20,22-dihydroxycholesterol.     

Requirement for radiation shields of transportation pipe for on line inhalation gases from compact cyclotron in positron emission tomography

In the unit housing of a compact cyclotron and positron emission CT (PET), positron emitting gas such as 15O, 11C, C15O2, C15O etc. is supplied from a cyclotron to a PET room through a transportation pipe with an appropriate shield to reduce positron annihilation radiation. Using lead or concrete shield blocks with various thicknesses, radiation leakage through the shield was measured by an ionization chamber type survey meter during continuous and constant supply of 15O gas of 1.85 GBq/min concentration which is the maximum dose for clinical use. The leakage radiation measured was 213.7, 56.0, 15.3, 5.0 muSv/week for lead shield with 1, 2, 3, and 4 cm thickness, respectively, and 193.3, 30.5 and 5.1 muSv/week for concrete shields with thickness of 10, 20, and 30 cm, respectively. The present study shows that to keep less than 300 muSv/week, which is the permissible dose rate of the boundary zone around the radiation controlled area by Japan Science and Technology Agency, it is required to use more than 8 mm thick lead shield or 7 cm thick concrete for continuous supply of 1.85 GBq/min 15O gas.     

Effect of brain lesions on [3H]ohmefentanyl binding site densities in the rat striatum and substantia nigra.

We have recently demonstrated that [3H]ohmefentanyl, a non-peptidergic opioid ligand which was suggested to cross the blood brain barrier in contrast to other peptidergic opioid ligands, bound not only to mu opioid receptor sites but also to sigma sites. In order to examine whether [3H]ohmefentanyl can be used as a marker for mu sites, we investigated the effects of brain lesions on [3H]ohmefentanyl binding site densities, as compared with [3H][D-Ala2, MePhe4, Gly-ol5]enkephalin ([3H]DAGO), a selective mu ligand. These binding site densities were measured by quantitative autoradiography in the rat striatum and substantia nigra, two brain structures known to contain a high density of mu receptors, following lesions of the nigro-striatal dopaminergic pathway and striatal intrinsic neurons. Following unilateral nigral lesion with 6-hydroxydopamine, [3H]ohmefentanyl binding site densities were decreased in the patches (-35%) and matrix (-20%) of the ipsilateral striatum and in the lesioned substantia nigra pars compacta (-49%). Unilateral striatal lesion with quinolinic acid induced 72%, 61% and 50% decreases in [3H]ohmefentanyl binding in the patches and matrix of the lesioned striatum and in the ipsilateral substantia nigra pars reticulata, respectively. Similar results were obtained in the binding of [3H]DAGO. Indeed, a significant linear correlation was observed between [3H]ohmefentanyl and [3H]DAGO binding site densities. Therefore, mu opioid receptors may be mainly located on intrinsic neurons in the striatum, dopaminergic cell bodies in the substantia nigra pars compacta and nerve terminals of striatal efferents in the substantia nigra pars reticulata.(ABSTRACT TRUNCATED AT 250 WORDS)     

Quartz crystal microbalance immunosensor for highly sensitive 2,3,7,8-tetrachlorodibenzo-p-dioxin detection in fly ash from municipal solid waste incinerators

A quartz crystal microbalance (QCM) immunosensor was developed for the detection of 2,3,7,8-tetrachlorodibenzo-p-dioxins (TCDD) in environmental pollutants. An anti-TCDD antibody was immobilized on the gold surface of the QCM via chemical coupling, and its immunologic activity was then maintained by treatment with an artificial stabilizing reagent such as poly(2-methacryloyloxyethyl phosphorylcholine-co-n-butyl methacrylate). A competitive immunoreaction with TCDD conjugated ovalbumin (TCDD-ovalbumin) was used to detect TCDD. A calibration curve was obtained through the competitive immunoreaction, and linearity was shown from 100 ng mL(-1) to 0.1 ng mL(-1). Also, the cross-reactivities of the anti-TCDD monoclonal antibody were thoroughly evaluated with several TCDD derivatives. The relationships between GC-MS, ELISA, and QCM were compared using fly ash samples from a municipal solid waste, which were prepared using an accelerated solvent extractor. For 23 samples, the experimental relationship between the TCDD concentration by QCM vs. the TCDD concentration by ELISA was y= 1.07x + 2.70, r= 0.99, and the TCDD concentration by QCM vs. the toxic equivalent quantity (TEQ) value by GC-MS was y= 2.46x - 14.98, r= 0.89.     

Analytical use of a biochemical luminous membrane

Luminous membranes were prepared by immobilizing peroxidase (POD) to collagen matrix. The POD luminous membrane generated luninescence in the presence of luminol and H₂O₂, and the peroxide was determined in the concentration range 10-⁶-10-³ M by following luminescence emitted from the membrane. Glucose was determined using a luminous membrane in which POD and glucose oxidase (GOD) were coimmobilized. The luminous membranes appear to be feasible for the determination of enzyme substrates and enzyme activity.
